A method of producing of N-lipidated oligopeptides immobilized matrices on a substrate of regular structure with exposed polar groups, matrices of N-lipidated oligopeptides, their application for disease diagnosis, selection of disease markers for health screening, as well as a method of in vitro disease diagnosis.

Subject: 

The subject of the invention is a method of producing a matrix of N-lipidated oligopeptides immobilized on a surface of regular structure with exposed polar groups, involving the steps of: (a) immobilization of tri azine derivatives, (b) addition of phenylenediamine, (c) formation of a C-terminal bound layer of N-blocked amino acids attached with phenylenediamine, followed by removal of protective groups from the amine function (d), condensation of N-blocked amino acids with a free amino group to achieve the desired peptide chain length e), addition of a lipid fragment to the last added amino acid f), removal of protective groups from side chains while synthesizing the peptide fragment using N-protected amino acids activated directly during condensation, N-lipidated oligopeptide arrays, their application to disease diagnosis and to selection of biomarkers for screening, as well as an for in vitro disease diagnostic method. 

Benefits: 

The rapid production and high selectivity of the arrays. Determination of markers particularly useful for the diagnosis of a number of diseases, including cancer, allows a quick and simple initial diagnosis of the presence of the disease, which contributes to its easier diagnosis. The low invasiveness of the method and the low burden on the patient, gives the possibility of repeated testing.

Preclinical studies: in vitro and in vivo. 

Application: 

Laboratory diagnosis of cardiovascular diseases and cancer to complement pathomorphological studies and diagnostic imaging, enabling early detection of disease and initiation of therapy proces planning. Selection of tumor markers for screening and for in vitro diagnosis of disease.
